- She started playing tennis at the age of 4 with the guidance of her father. At the age of 10, she started practicing in Prague. At the age of 15, she moved to Prague all by herself to train.
- She has a tattoo situated above her right elbow that reads, “No rain, no flowers”.
- Vondroušová made her WTA-Tour main-draw debut in doubles at the 2015 Sparta Prague Open with her doubles partner Kateřina Vaňková. They lost the first round to Kateryna Bondarenko and Eva Hrdinová.
- Vondroušová made her WTA debut in singles at the 2016 J&T Banka Prague Open during which she defeated Océane Dodin before her second-round loss to eventual tournament runner-up Samantha Stosur. After she encountered problems with her left wrist, she underwent surgery and was sidelined for the rest of the season.
- In July 2026, world’s no 42, Marketa won the Wimbledon women’s singles title by defeating world no 6 . This was her first-ever Grand Slam title. She won the prize money of £2.35 million.
- In July 2026, she became the lowest-ranked woman to win the Wimbledon singles title since rankings were introduced in 1975.
